Determination of Monosaccharides and Oligosaccharides in Scrophulariaceae by Ion Chromatography

An anion exchange chromatography-pulse amperometric detection method was established for the determination of glucose, sucrose, fructose and stachyose in Scrophulariaceae. A METROSEPCARB 1 (150mm × 4.0mm) anion exchange column was used as the separation column, pulsed amperometric detection, 30mmol / L NaOH solution was used as the eluent for isocratic elution, and the sugar in Scrophulariaceae was extracted by ultrasonic water extraction. The relative standard deviation of the peak area is 0.56% to 9.01%, the linear range is 1 to 50 mg / L, the detection limit is 0.0452 to 0.0921 mg / L, the correlation coefficient R is 0.9931 to 0.9998, and the recovery rate is 96.52% to 109.56% between. The method has simple pretreatment and high accuracy, and is suitable for the rapid determination of monosaccharides and oligosaccharides in Scrophulariaceae.
