Rat β amyloid 1-40 (Aβ1-40) enzyme-linked immunoassay kit instruction manual

This reagent is intended for research use only and is designed to quantify the levels of amyloid beta 1-40 (Aβ1-40) in rat serum, plasma, and other biological liquid samples. The assay is based on the double-antibody sandwich immunoassay principle. A microtiter plate is pre-coated with a specific monoclonal antibody against rat Aβ1-40, which acts as a solid-phase capture antibody. After incubation with the sample, the target Aβ1-40 binds to the immobilized antibody. Subsequently, a horseradish peroxidase (HRP)-labeled secondary antibody is added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing to remove unbound components, the TMB substrate is introduced. Under the catalytic action of HRP, TMB turns blue and then changes to yellow when an acidic stop solution is added. The intensity of the color developed is directly proportional to the concentration of Aβ1-40 in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the concentration of Aβ1-40 in the sample is determined by comparing it to a standard curve generated from known concentrations. The kit includes all necessary reagents for performing the assay. It is available in both 48-well and 96-well configurations.
