This reagent is intended for research use only. The kit is designed to quantitatively determine the levels of amyloid beta 1-40 (Aβ1-40) in rat serum, plasma, and other related biological samples. The assay is based on the double-antibody sandwich immunoassay principle. A microplate is pre-coated with a purified monoclonal antibody specific to rat Aβ1-40, forming a solid-phase antibody. After adding the sample, the target Aβ1-40 binds to the immobilized antibody. Subsequently, an HRP-conjugated anti-Aβ1-40 antibody is added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing to remove unbound components, a TMB substrate solution is introduced. Under the catalytic action of HRP, TMB turns blue and then changes to yellow upon addition of an acidic stop solution. The intensity of the color developed is directly proportional to the concentration of Aβ1-40 in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the concentration of Aβ1-40 in the sample is determined by comparing the OD value with a standard curve.
