Human papillomavirus (HPV) enzyme-linked immunosorbent assay (ELISA) kit instruction manual

This ELISA kit is specifically designed for the qualitative detection of Human Papillomavirus (HPV) in various biological samples, including human serum, plasma, tissue, cell culture supernatants, and other related fluids. It is intended for research use only and not for diagnostic or clinical purposes. The shelf life of the kit is 6 months when stored properly at 2–8°C. The assay is based on a double-antibody sandwich ELISA method. A microtiter plate is pre-coated with purified HPV-specific antibodies, acting as a solid-phase capture reagent. When the sample is added, any HPV present will bind to these immobilized antibodies. After washing to remove unbound substances, a horseradish peroxidase (HRP)-labeled HPV antibody is introduced, forming a complex of antibody-antigen-enzyme-labeled antibody. Following another wash step, TMB substrate is added, which turns blue in the presence of HRP. The reaction is then stopped by adding an acidic solution, causing the color to change to yellow. The optical density (OD) is measured at 450 nm using a microplate reader, and results are interpreted against a predefined cutoff value to determine the presence or absence of HPV in the sample. The kit includes all necessary reagents for performing the assay. It comes in two configurations: 48-well and 96-well formats. The components include: - RT Sealing Plate (2 pieces for 48-well, 2 pieces for 96-well) - RT Sealed Bag (1 piece each) - Enzyme-Linked Immunoassay Plate (1×48 or 1×96, stored at 2–8°C) - Negative Control (0.5 ml × 1 bottle, stored at 2–8°C) - Positive Control (0.5 ml × 1 bottle, stored at 2–8°C) - Enzyme Standard Reagent (3 ml × 1 bottle or 6 ml × 1 bottle, stored at 2–8°C) - Sample Diluent (3 ml × 1 bottle or 6 ml × 1 bottle, stored at 2–8°C) - Developer A (3 ml × 1 bottle or 6 ml × 1 bottle, stored at 2–8°C) - Color Reagent B (3 ml × 1 bottle or 6 ml × 1 bottle, stored at 2–8°C) - Stop Solution (3 ml × 1 bottle or 6 ml × 1 bottle, stored at 2–8°C) - Concentrated Wash Solution (20 ml × 20 times or 20 ml × 30 times, stored at 2–8°C) Proper storage and handling are essential to ensure accurate and reliable results. Always refer to the complete instructions provided with the kit for detailed procedures and safety information.
