This is the user manual for the Human Papillomavirus (HPV) Enzyme-Linked Immunosorbent Assay (ELISA) Kit. Please note that this kit is intended for research use only and not for diagnostic or clinical purposes. It is designed to quantitatively detect the presence of human papillomavirus in various biological samples, including human serum, plasma, tissue, cell culture supernatants, and other related fluids. The shelf life of the kit is 6 months when stored properly. Storage conditions: Keep the kit at a temperature of 2–8°C. Avoid freezing unless specified otherwise. Principle of Operation: This ELISA kit utilizes a double-antibody sandwich method to detect HPV antigens in the sample. A microtiter plate is pre-coated with purified anti-HPV antibodies, forming a solid-phase capture antibody. When the sample is added, any HPV present will bind to these antibodies. Unbound materials are then washed away. A horseradish peroxidase (HRP)-labeled anti-HPV antibody is added, forming a complex of antibody-antigen-enzyme-labeled antibody. After washing, the substrate TMB (3,3',5,5'-tetramethylbenzidine) is introduced. Under the catalytic action of HRP, TMB turns blue and is then stopped by adding an acidic solution, resulting in a yellow color. The optical density (OD) at 450 nm is measured using a microplate reader. The result is compared against a pre-determined cutoff value to determine whether HPV is present in the sample. Kit Components: - 48-well configuration: 1 plate, 2 RT sealing plates, 1 RT sealed bag, 1 enzyme-labeled plate, 1 bottle each of Negative Control, Positive Control, Enzyme Standard Reagent, Sample Diluent, Developer A, Developer B, and 1 concentrated wash solution (20ml × 20 times). - 96-well configuration: 1 plate, 2 RT sealing plates, 1 RT sealed bag, 1 enzyme-labeled plate, 1 bottle each of Negative Control, Positive Control, Enzyme Standard Reagent, Sample Diluent, Developer A, Developer B, and 1 concentrated wash solution (20ml × 30 times). All components should be stored at 2–8°C until use. Ensure all reagents are thoroughly mixed before use and follow the instructions carefully to obtain accurate results. Do not mix reagents from different batches. Always use fresh reagents and avoid contamination.
