The Composition of Media

Medium and Ingredients

1. Acetobacter Medium

Glucose (glucose) 100g, Yeast extract (yeast extract) 10g, CaCO3 20g, Agar (agar) 15g, Distilled water 1000ml, Adjust pH to 6.8.

Scope of application: Odorous variant of Acetobacter aceti.

2. Nutrient Agar (Nutrition Gravy Agar)

Peptone 5g, Beef extract 30g, NaCl 5g, Agar (agar) 15g, Distilled water, Adjust pH to 7.0-7.2.

Note: When cultivating Bacillus, 5mg of MnSO4·H2O may be added for better spore formation.

Scope of application: Gas-producing bacteria, Alcaligenes faecalis, Bacillus cereus, Bacillus licheniformis, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Saccharomyces cerevisiae, and more.

3. Corn Meal Medium

Maize flour 5g, Peptone 0.1g, Glucose (glucose) 1g, Tap water 1000ml.

Note: Boil the mixture in an autoclave at 121°C for 1 hour, then distribute into 18x18 mm tubes with 10 ml each. Autoclave again at 121°C for 1 hour.

4. Lactic-bacteria Medium I (Lactic Acid Bacteria Medium I)

Yeast extract 7.5g, Peptone 7.5g, Glucose (glucose) 10g, KH2PO4 2g, Tomato juice 100ml, Tween 80 0.5ml, Distilled water 900ml, pH 7.0.

Scope of application: Lactobacillus plantarum, thermophilic lactic acid streptococci.

5. Lactic-bacteria Medium II (Lactic Acid Bacteria Medium II)

Lacto-casein peptone 10g, Beef extract 10g, Yeast extract 5g, Glucose (glucose) 5g, Tween 80 1g, K2HPO4 2g, Na-acetate 5g, Diamine citrate 2g, MgSO4·7H2O 0.2g, MnSO4·H2O 0.05g, Distilled water 1000ml, pH 6.5–6.8.

Scope of application: Lactobacillus plantarum.

6. Peptone Glucose Yeast extract Medium PGY

Peptone 10g, Yeast extract 5g, Glucose (glucose) 1g, Distilled water 1L.

7. Glycerol Agar

Peptone 5g, Beef extract 3g, Glycerol (Glycerin) 20g, Tap water 1000ml, Agar (agar) 15g, pH 7.0–7.2.

8. Rhizobium medium AS 9

Yeast extract 1g, Soil extract 200ml, Mannitol 10g, Agar (agar) 15g, Distilled water 800ml, pH 7.2.

Note: Prepare soil extract by suspending 50g dried garden soil in 200ml tap water, boil, filter, and adjust volume to 200ml.

Scope of application: Soybean rhizobium, cowpea rhizobium, peanut rhizobium, and others.

9. Mannitol Agar

Yeast extract 5g, Peptone 3g, Mannitol 25g, Agar (agar) 15g, Distilled water 1000ml.

10. Glucose Asparagine Agar

Glucose (glucose) 10g, Asparagine (asparagine) 0.5g, K2HPO4 0.5g, Water 1000ml, pH 7.2–7.4.

Scope of application: Pseudomonas aeruginosa, Micrococcus roseus.

11. Gause's Synthetic Agar (Gao's Synthetic No. 1 Agar)

KNO3 1g, Soluble starch 20g, K2HPO4 0.5g, MgSO4·7H2O 0.5g, NaCl 0.5g, FeSO4 0.01g, Agar (agar) 20g, Water 1000ml, pH 7.2–7.4.

Scope of application: Pseudomonas aeruginosa, Streptomyces species, and more.

12. Wort Agar

Dilute wort (without hops) to 12 Brix, add 15g agar per 1000ml, melt, and dispense into tubes. Autoclave at 110°C for 30 minutes.

Scope of application: Candida, Geotrichum, Aspergillus, Saccharomyces, and many yeast strains.

13. Potato Dextrose Agar (PDA)

Potato extract 1000ml, Dextrose (glucose) 20g, Agar (agar) 20g.

Note: Prepare potato extract by boiling peeled potatoes in water, filtering, and adding glucose and agar.

Scope of application: Penicillium, Aspergillus, Fusarium, Mucor, and many fungi.

14. Czapek’s Agar

Sucrose (sucrose) 30g, NaNO3 3g, MgSO4·7H2O 0.5g, KCl 0.5g, FeSO4·7H2O 0.01g, K2HPO4 1g, Agar (agar) 13g, Distilled water 1000ml, pH 7.2.

Scope of application: Aspergillus, Penicillium, Streptomyces, and other filamentous fungi.

15. Concentrated Sucrose Czapek’s Agar

Sucrose (sucrose) 200g, NaNO3 3g, MgSO4·7H2O 0.5g, KCl 0.5g, FeSO4·4H2O 0.01g, K2HPO4 1g, Agar (agar) 13g, Distilled water 1000ml.

Scope of application: Aspergillus species.

16. Synthetic Potato Medium

20% Potato extract 1L, Glucose (glucose) 20g, KH2PO4 3g, MgSO4·7H2O 1.5g, Thiamine (B1) trace, Agar (agar) 15g, pH 6.

Scope of application: Aspergillus, Penicillium, Fusarium, Trichoderma, and many fungi.

17. Corn Meal Medium

Mix corn meal with water (20:100 W/V), fill into 100ml bottles with 30ml each, autoclave at 121°C for 30 minutes.

18. Filter Paper Medium

(NH4)2SO4 1g, KH2PO4 1g, MgSO4·7H2O 0.7g, NaCl 0.5g, Distilled water 1000ml, 6x1cm filter paper strip.

19. Soybean Cake Meal Agar

10% soybean cake infusion, 1000ml, Glucose 10g, NaCl 2.5g.

20. Sawdust, Wheat Bran Medium

Broadleaf sawdust 75g, wheat bran 25g, mix well, add water, fill into bottles, plug with cotton, sterilize at 15 lbs for 1 hour.

21. Pine Block or Pine Sawdust Medium

Soak pine blocks in 1–2% sucrose solution, fill into bottles, wrap in paper, sterilize at 20 lbs for 1 hour.

22. Rice Straw-Dung Medium

Dried horse dung 65%, rice straw 35%, urea 0.5%, superphosphate 0.7%, gypsum 1%, ammonium sulfate 0.3%.

Note: Add water, pile, and allow fermentation.

23. Straw-Rice Bran Medium

Dried straw 75–80%, rice bran 20–25%, soak in water for 12 hours, drain, mix, and sterilize.

24. Rice Medium

Fill 10g rice into a 50ml flask, add 20–30ml water, autoclave at 121°C for 30 minutes.

25. Filter Paper Medium

(NH4)2SO4 1g, KH2PO4 1g, MgSO4·7H2O 0.5g, Yeast extract 0.1g, Distilled water 1000ml, 1x7cm filter paper strip.

Note: Distribute 1ml per tube, sterilize, then add filter paper aseptically.

26. Yeast Extract-Malt Extract Agar I

Yeast extract 10g, Malt extract 10g, Glucose 4g, Distilled water 1000ml, Agar 20g, pH 7.0.

Scope of application: Streptomyces erythromycin.

27. Yeast Extract Peptone Agar

Yeast extract 1g, Multi-peptone 2g, Beef extract 1g, Glucose 10g, Agar 20g, Distilled water 1L, pH 7.0.

28. Glucose Yeast Extract Starch Agar

Glucose 10g, Yeast extract 10g, Starch 10g, NaCl 5g, CaCO3 3g, Agar 20g, Distilled water 1L, pH 7.0.

29. Wheat Bran Medium

Wheat bran 36g, (NH4)2HPO4 10g, K2HPO4 0.2g, MgSO4·7H2O 0.1g, Agar 20g, Distilled water 1L, pH 7.0.

30. Yeast Sporulation Medium

1. Kleyn Medium: KH2PO4 0.012%, K2HPO4 0.02%, Sodium acetate 0.5%, Glucose 0.062%, NaCl 0.062%, Peptone 0.25%, Biotin 2μg%, Agar 2%, Trace element solution 1ml/100ml.

2. Gorodkowa Medium: Glucose 0.1%, Peptone 1%, NaCl 0.5%, Agar 2%.

3. McClary Medium: Glucose 0.1%, KCl 0.18%, Yeast juice 0.25%, Sodium acetate 0.82%, Agar 1.5%.

31. BPY Medium

Beef extract 5g, Peptone 10g, NaCl 5g, Yeast extract 5g, Glucose 5g, Agar 1–2%, Distilled water 1000ml, pH 7.0.

Scope of application: Escherichia coli, Pseudomonas putida.

32. LB Medium

Yeast extract 5g, Peptone 10g, NaCl 10g, Agar 1–2%, Distilled water 1000ml, pH 7.0.

Scope of application: Escherichia coli.

33. Penssay Broth

Beef extract 1.5g, Yeast extract 1.5g, Tryptone 5g, NaCl 3.5g, Glucose 1g, K2HPO4·3H2O 4.8g, Distilled water 1000ml, pH 7.2.

34. Blood Agar

Peptone 10g, Glucose 2g, NaCl 5g, NaH2PO4 2.5g, Sodium citrate 5g, Brain infusion 800ml, Pig heart infusion 200ml, 10% blood, pH 7.2.

Note: Prepare pig brain and heart infusions by boiling, filtering, and combining.

35. Cellulose-decomposing Bacteria Synthetic Medium

NaCl 6g, MgSO4·7H2O 0.1g, KH2PO4 0.5g, CaCl2 0.1g, K2HPO4 2.0g, Yeast extract 1.0g, (NH4)2SO4 2.0g, Distilled water 1000ml, Filter paper strip (1 piece), pH 7.0–7.4.

Note: For cellulose-degrading bacteria, use 2% carboxymethyl cellulose (CMCNa) and 2% agar.

36. Clostridium Pasteurianum Synthetic Medium

Glucose 1%, MnSO4 0.001%, KH2PO4 0.05%, FeSO4 0.001%, K2HPO4 0.05%, Yeast extract 0.1%, MgSO4·7H2O 0.02%, Peptone 0.01%, NaCl 0.001%, CaCO3 0.5%, Distilled water 100%, pH 7.0.

37. Yeast Extract-Malt Extract Agar II

Yeast extract 4g, Malt extract 10g, Glucose 4g, Agar 17g, Distilled water 1000ml, pH 7.3.

38. Oatmeal Agar

Oatmeal (infusion) 20g, Agar 18%, Trace element solution 1ml, Distilled water 1000ml, pH 7.2.

Note: Trace element solution: FeSO4·7H2O 0.1g, MnCl2·4H2O 0.1g, ZnSO4·7H2O 0.1g, Distilled water 100ml.

39. Calcium Malate Agar

Calcium malate 10g, K2HPO4 0.5g, NH4Cl 0.5g, Glycerol 10g, Agar 15g, Distilled water 1000ml.

40. Thiobacillus Ferrooxidans Medium

(NH4)2SO4 0.15g, KH2PO4 0.05g, KCl 0.05g, MgSO4·7H2O 0.5g, Ca(NO3)2·4H2O 0.01g, FeSO4·7H2O 50g, Distilled water 1000ml, pH 2.0.

41. Thiobacillus Thiooxidans Medium

(NH4)2SO4 0.3g, KH2PO4 3–4g, CaCl2 (anhydrous) 0.25g, MgSO4·7H2O 0.5g, FeSO4·7H2O 0.001g, Distilled water 1000ml, pH 3.5–4.

42. Plasma Substitute Medium

Sucrose 12.5–15%, Peptone 0.5%, KH2PO4 0.03%, Na2HPO4·12H2O 0.14%, pH 7.0–7.2.

43. Skim Milk Medium ATCC 183

Skim milk 100g, deionized water 1000ml.

Scope of application: Lactobacillus bulgaricus, Streptococcus thermophilus.

44. Pea Agar

Glucose 1%, Peptone 0.5%, NaCl 0.5%, Agar 2.0%, Pea infusion (counted by NH2) 12–14 mg/100ml, pH 7.2–7.5 before sterilization.

Note: Prepare pea infusion by soaking peas in water, adjusting pH, adding chloroform, and fermenting for 7–10 days.

Standard quality: Aminonitrogen 160–200 mg/100ml, soluble phosphorus 400–600 μg/ml, golden and transparent.
