What is the difference between using fetal bovine serum and calf serum?
If fetal bovine serum (FBS) and calf serum (CS) are both available, many research biocell growers tend to prefer fetal bovine serum. But why is that? What really sets FBS apart from CS? Let’s take a closer look at the differences and why FBS might be the better choice in many cases.
First, the source of the serum is different. Fetal Bovine Serum is obtained by cardiac puncture from a fetus inside a pregnant cow at the time of slaughter. This means it comes from a very young animal, typically around 6-8 months old. On the other hand, Calf Serum is collected from calves that are just a few days to two weeks old, right after birth. The age difference plays a significant role in the quality and composition of the serum.
Second, the composition and proportions of the active components differ significantly. FBS contains a broader range of growth factors, adhesion-promoting substances, and hormones, which can support a wider variety of cell types. Some cells, like stem cells or certain primary cultures, require the specific growth factors found only in FBS. In contrast, some cell lines may thrive perfectly well with calf serum. The concentration also varies—typically between 5% and 10%, though some specialized applications may require up to 20%. Choosing the right serum depends on the cell type and the experiment's needs.
Third, when considering overall quality, FBS has an advantage. Since fetal cows live in a controlled and clean environment, their serum is less likely to be contaminated compared to serum from older calves. If the production process is strictly controlled and standardized, FBS tends to offer more consistent performance. That’s why many labs and researchers prefer FBS for sensitive experiments. However, for most general cell culture applications, calf serum can still be a cost-effective and reliable option.
